Western blot was performed using Anti-Desmin Recombinant Rabbit Monoclonal Antibody (SI18-00) (Product # MA5-32068) and bands between 45-55 kDa corresponding to Desmin were observed in RD cell lysate but not in MCF7 cell lysate, and increased in expression in iPSCs upon differentiation to cardiomyocytes. Whole cell extracts (30 µg lysate) of RD (Lane 1), MCF7 (Lane 2), iPSC (Lane 3), iPSC differentiated to cardiomyocytes (Lane 4), and tissue extract (30 µg lysate) of mouse colon (Lane 5) were electrophoresed using NuPAGE™ 10% Bis-Tris Protein Gel (Product # NP0302BOX), 12 well. Resolved proteins were then transferred onto a nitrocellulose membrane (Product # IB23001) by iBlot® 2 Dry Blotting System (Product # IB21001). The blot was probed with the primary antibody (1:2000 dilution) and detected by chemiluminescence with Goat anti-Rabbit IgG (Heavy Chain) Superclonal™ Recombinant Secondary Antibody, HRP (Product # A27036, 1:10000) using the iBright™ FL1500 Imaging System (Product # A44115). Chemiluminescent detection was performed using SuperSignal™ West Pico PLUS Chemiluminescent Substrate (Product # 34580).
